摘要
转录因子FoxO1是叉头家族O的亚族成员之一,是胰岛素/胰岛素样生长因子信号通路中的关键分子。FoxO1蛋白主要有磷酸化、乙酰化和泛素化3种不同形式的共价修饰,这3种修饰方式在调节FoxO1蛋白分子转录活性和信号转导过程中发挥着重要的作用。FoxO1在胰岛β细胞中表达丰富,与其生长增殖、凋亡、分化、应对氧化应激等密切相关。FoxO1作为胰岛β细胞中特异的转录因子,研究其在β细胞中的表达及作用,将为糖尿病的预防和诊治带来新的契机。
FoxO1 ,playing a key role in INS/IGF-1 ( insulin/insulin like growth factor 1 )signaling pathway,is one of the O subgroup members in Forkhead family. There are three various forms of post-translation modifications,namely phosphorylation, acetylation and ubiquitination. These modifications contribute to the process of signal transduction and are involved in the regulation of FoxO1 transcriptional activity. FoxO1 is expressed in pancreatic β-cells abundantly,with a close relationship with its proliferation, apoptosis,differentiation,and response to oxidative stress. The research on FoxO1, a specific transcription factor in pancreatic β-cells, will bring a new chance in the prevention, diagnosis, and treatment of diabetes mellitus.
