摘要
目的:比较不明原因复发性流产(URSA)与正常早孕蜕膜CD8和颗粒酶B的表达,从蛋白水平探讨不明原因复发性流产的发病机制。方法:用免疫组织化学方法测定两组蜕膜CD8和颗粒酶B蛋白表达,采用双盲法随机取5个视野,400倍下观察蜕膜组织处阳性细胞,计数后取其平均数进行半定量分析。结果:与正常早孕相比,URSA组蜕膜组织处颗粒酶B表达明显升高(P<0.01);CD8的表达两组比较无明显差异(P>0.05)。结论:早孕期蜕膜组织颗粒酶B蛋白表达异常是URSA的机制之一;颗粒酶B死亡途径可能是早期诱导URSA发生的重要因素。
Objective:To compare the expressions of granzyme B and CD8 in decidual tissues of cases with unexplained recurrent spontaneous abortion(URSA) and normal early pregnant women,explore the mechanism of URSA according to protein level.Methods:Immunohistochemical method was used to detect the expressions of granzyme B and CD8 in decidual tissues of cases with URSA,5 visual fields were selected randomly by double-blind method,positive cells in decidual tissues were observed under microscope(400×),mean was obtained after counting for semiquantitative analysis.Results:Compared with normal early pregnancy group,the expression level of granzyme B in decidual tissues of URSA group increased significantly(P0.01);but there was no significant difference in expression level of CD8 between the two groups(P0.05).Conclusion:The abnormal expression of granzyme B protein in decidual tissues of URSA group is one of the mechanisms of URSA;the pathway of cell death induced by granzyme B may be an important factor of occurrence of URSA.
