非小细胞肺癌患者外周血中淋巴细胞肝素酶基因表达与淋巴结转移的关系

Relationship between the expression of heparanase gene in peripheral blood lymphocytes and lymph nodes metastasis in patients with non-small cell lung cancer

摘要 目的探讨非小细胞肺癌(NSCLC)患者外周血中淋巴细胞肝素酶基因表达与淋巴结转移的关系。方法采用逆转录-聚合酶链反应(RT-PCR)技术对62例NSCLC患者手术前、后外周血及15例正常人外周血中淋巴细胞肝素酶表达水平进行检测。结果 28例淋巴结转移患者肝素酶基因阳性表达率89.29%(25/28),34例无淋巴结转移的患者肝素酶基因阳性表达率89.29%(25/28),差异有统计学意义(P<0.01),而15例正常人外周血中淋巴细胞肝素酶均呈阴性表达。25例有淋巴结转移的肝素酶基因阳性NSCLC患者手术前、后外周血中淋巴细胞肝素酶基因表达水平高于10例无淋巴结转移的肝素酶基因表达水平,差异有统计学意义(P<0.01);而手术前后外周血中淋巴细胞肝素酶基因表达水平差异无统计学意义(P>0.05),鳞癌患者手术前后外周血中淋巴细胞肝素酶基因阳性表达率低于腺癌患者,差异有统计学意义(P<0.05)。结论 NSCLC患者外周血中淋巴细胞肝素酶基因表达水平与非小细胞肺癌淋巴结转移有关,它可能成为预测非小细胞肺癌转移与否的指标。 Objective To investigate the relationship between the expression of heparanase gene in peripheral blood lymphocytes and lymph nodes metastasis in patients with non-small cell lung cancer.Methods The expression of heparanase gene in peripheral blood lymphocytes from 62 patients with non-small cell lung cancer was detected by reverse transcriptase-polymerase chain reaction(RT-PCR),and 15 healthy subjects served as normal controls.Results The expression of heparanase gene in patients with lymph node metastasis was significantly higher than those without lymph node metastasis(P〈0.01).There was no significant difference in the expression before and after operation(P〉0.05).The expression of heparanase in normal control group was negative.The expression of heparanase gene in patients with squamous carcinoma was obviously lower than those with adenocarcinoma(P〈0.05).Conclusion The expression of heparanase gene in peripheral blood lymphocytes may be related with the metastasis of non-small cell lung cancer,which may be used as a prognostic marker for the metastasis of non-small cell lung cancer.
