摘要
目的:探讨抗凋亡蛋白XIAP和促凋亡蛋白Smac在儿童急性白血病(AL)中的表达及临床意义。方法:应用SP-免疫组织化学方法检测60例AL患儿初治组、缓解组、复发组及25例非恶性血液病患儿(对照组)中XIAP、Smac的表达情况。结果:1 XIAP、Smac在初治AL患儿骨髓中表达阳性率及阳性强度均高于缓解组(P<0.05,P<0.008)和对照组(P<0.05,P<0.008),与复发组相比无显著性差异(P>0.05)。2初治AL患儿骨髓中XIAP、Smac蛋白表达显著正相关(r=0.795,P<0.01)。结论:XIAP、Smac过度高表达和急性白血病的发病和复发相关,XIAP、Smac在儿童AL的发生与发展中可能互相影响。
Objective: To investigate the expression and their clinical significance of anti-apoptotic protein XIAP and pro-apoptotic protein Smac in the children with AL(Acute leukemia).Methods: The expression of XIAP and Smac were measured by immunohistochemical assay in 60 children with AL,including 30 newly diagnosed and untreated AL children,26 children in remission and 4 relapsed children.Other 25 children without malignant blood disease and with normal bone marrow were selected as normal controls(NC).Results: ① The positive rate and expression intensity of XIAP and Smac in the bone marrow of newly diagnosed AL children were all higher than those in remission and normal controls(P 0.05,P 0.008),compared with those in relapsed children,the difference have no statistical significance(P 0.05).②The protein level of XIAP in newly diagnosed AL children was positively correlated with Smac(r=0.795,P0.01).Conclusion: Over expression of XIAP and Smac may play synergic roles in the pathogenesis of leukemia and higher risk of relapse.XIAP and Smac may affect each other in the genesis and progression of AL in children.
