摘要
为构建含有I型鸭病毒性肝炎病毒3D基因的重组鸭瘟病毒,本研究将已建立的鸭瘟病毒TK基因缺失转移载体(pBl ueSK-TK-EGFP)进行改造,在其荧光表达盒内插入I型鸭病毒性肝炎病毒3D基因,将重组后的转移载体(pBl ueSK-TK-EGFP-3D)转染已感染鸭瘟病毒的鸭胚成纤维细胞,转染细胞盲传2代后仍能观察到绿色荧光。表明本研究已成功构建携带I型鸭病毒性肝炎病毒3D基因的重组鸭瘟病载体(pBl ueSK-TK-EGFP-3D),此研究为研制鸭瘟-鸭肝炎二联苗奠定了基础。
In order to construct recombinant Duck enteritis virus(DEV) containing 3D gene of Duck hepatitis virus type I(DHV-I),the 3D gene of DHV-I was amplified and inserted into EGFP expression cassette of TK gene-deleted transfer vector(pBlueSK-TK-EGFP) for DEV.The recombinant transfer vector(pBlueSK-TK-EGFP-3D) was transfected into the DEV infected duck embryo fibroblasts(DEF) and the green fluorescence was still observed in cells after the second blind passage.These results showed that transfer vector pBlueSK-TK-EGFP-3D had been constructed successfully.This study laid the foundation for development of a bivalent vaccine against DEV and DHV-I.
