摘要
对小麦幼穗的21个基因型、3个发育时期、6种培养基在幼穗离体培养中的反应进行比较和筛选。建立了繁殖力和分化力均较强并保持长达9个月的体细胞无性系。
The response of 3 development stages and 21 genotypes of young spike of wheat and 6 media to culture in vitro of young spike and other relative aspects were studied.The results have been obtained as follows:(1)There was a gre- at differene in the effects of young spike culture in vitro among the three differentiation stages:namely,the glume differentintion stage,the floret differen tiation stage and the pistil and stamen differentiation stage.The pistil and sta- men differentiation stage was the best,the floret the middle and the glume the worst.(2)The growth state and differention ability of young spike callus were strongly affected by the genotype.Most of 21 genotypes were bad,and four good genotypes e.g.Metipak 66,Jinghong-8,KC and ALD we(?)e selected.(3)Ad- ding BAP and CH or NAA could in hibit the growth of callus.The difference of v arious genotypes was comparatively stable on different media,but the affec of some genotypes could be regulated by the opponents of media.(4)The growth and differentiation ability of young spike callus of wheat were stable.But a very stable somaclone was established by the selection from Mexipak 66.
