摘要
目的 探讨水飞蓟素对肾间质纤维化大鼠血管紧张素Ⅱ(AngⅡ)表达的影响.方法 56只雄性SD大鼠完全随机分为假手术组8只,梗阻组24只,治疗组24只.梗阻组和治疗组大鼠利用单侧输尿管结扎法诱导肾小管间质纤维化模型,治疗组于治疗前30 min开始给予水飞蓟素30 mg/(kg·d)灌胃,梗阻组和假手术组同期给予生理盐水灌胃.梗阻组和治疗组分别于实验第7、14、21天各处死大鼠8只,假手术组于第21天处死全部大鼠,术侧肾组织观察病理变化并应用免疫组织化学方法检测肾组织AngⅡ的表达.结果 假手术组、梗阻组与治疗组第21天时肾小管间质评分为(0.14±0.09)、(2.49±0.26)、(1.89±0.24)分;AngⅡ的表达分别为(0.24±0.07)%、(12.26±0.18)%、(8.48±0.44)%;梗阻组与治疗组各指标明显高于假手术组,治疗组低于梗阻组.结论 水飞蓟素可能是通过下调AngⅡ表达而抑制纤维化的发生,从而起到对肾脏的保护作用.
Objective To investigate the effects of silymarin on the expression of Angiotensin Ⅱ in the kidneys of rats ith unilateral ureteral obstruction(UUO) -induced renal fibrosis. Methods Fifty-six rats were randomly assigned to shame peration group,operation group and silymarin[30 mg/(kg·d)] treatment group after unilateral ureteral obstruction. Tubulointerstitial fibrosis model was established via unilateral ureteral obstruction(UUO).Renal tissues were examined by light microscopy 7,14,21 days after operation. Immunhistochemistry was applied to measure the expression of Angiotensin Ⅱ in the renal interstitium. Results Silymarin may significantly ameliorate pathological change of nephridial tissue after unilateral ureteral obstruction. Silymarin may down-regulate the expression of Angiotensin Ⅱ in renal tissue. Conclusion Silymarin may inhibit renal fibrosis by down-regulating Angiotensin Ⅱ expression,which may play a key role in protection against renal fibrosis.
