三聚氰胺在奥尼罗非鱼体内的药动学研究 被引量:1

摘要 采用高效液相色谱串联质谱内标定量法,研究内服给药途径下,三聚氰胺在奥尼罗非鱼Oreochrom is aureus×O.niloticus体内的药动学特征.按奥尼罗非鱼体质量2.0 mg/kg的剂量内服三聚氰胺后,其血浆中药物浓度-时间曲线关系符合一级吸收一室模型.三聚氰胺在奥尼罗非鱼体内的主要药动学参数:吸收半衰期(t1/2ka)为(0.24±0.14)h;消除半衰期(t1/2ke)为14.33 h;达峰时间(tm ax)为1.42 h,峰浓度(Cm ax)为0.26 mg.L-1,平均滞留时间(MRT)为18.01 h.三聚氰胺在奥尼罗非鱼体内的主要药动学特征为:吸收迅速,达峰时间短,消除比较缓慢. Hybrid tilapias,Oreochromis aureus×O.niloticus were treated by oral administration of melamine in a single dose of 2.0 mg/kg.Melamine concentrations in hybrid tilapia plasma collected over 96 h following the treatment,were determined by HPLC-MS/MS,and pharmacokinetic parameters were analyzed using the computer program WinNonLin.It was shown that after a single dosage of oral administration,plasma melatonin concentration-time data could be described by one-compartment open model with first absorption.The main parameters were as follows:t1/2ka=(0.24±0.14)h,t1/2ke=14.33 h,tmax=1.42 h,Cmax=0.26 mg·L-1,MRT=18.01 h.These parameters indicated that melamine were rapidly absorbed,slowly eliminated in hybrid tilapias.
