葛根素衍生物4ac聚乳酸纳米粒制备工艺的初选及优化

摘要 目的筛选葛根素衍生物纳米粒的制备工艺条件。方法以纳米粒胶体溶液的外观、包封率、载药量为评价指标,通过单因素实验初选并经正交实验设计进行优化,确定最佳制备工艺条件。结果最佳工艺条件为PLA 100mg,药物30mg,25%卵磷脂0.2mL,F-68 350mg。按最佳工艺条件制备的葛根素衍生物纳米粒平均包封率为83.56%,载药量为22.32%。结论该研究确定的葛根素衍生物纳米粒的制备工艺条件稳定,可用于葛根素衍生物纳米粒的制备。 Objective To select the best prescription and preparation technology of 4ac-PLA-NP. Methods With embedding ratio and drug loading as indexes, 4ac-PLA-NP was prepared by a solvent evaporation method with PLA as carrier material, the technology was optimized by single factor test and orthogonal design test. Results In this optimum condition, a whole colloidal solution could be prepared in which embedding ratio was 83.56%, and drug loading was 22.32%. The optimum condition was PLA100 mg, 4ac 30 mg, 25% phospholipids 0~2 mL and F-68 350mg. Conclusion The optimum condition was stable and can be used for preparation of 4ac-PLA-NP.
