摘要
目的在体外探讨内皮抑素(ES)基因对骨肉瘤细胞生物学行为的影响。方法 ES基因重组质粒在体外扩增,应用脂质体法对骨肉瘤细胞株UMR106进行转染,Zeocin抗性筛选,MTT法观察ES基因对UM106细胞增殖能力的影响,ELISA法检测转染后细胞的ES分泌情况,Transwell小室观察细胞的侵袭及运动能力改变情况。结果转染后的UMR106细胞增殖能力与未转染相当,可分泌ES,浓度达350ng/ml,在体外转染后的细胞侵袭及运动能力无明显改变。结论应用ES基因在体外可成功转染骨肉瘤细胞,而且其对骨肉瘤细胞的增殖、运动及侵袭能力均无影响。
Objective To investigate the effect of endostatin gene on the biological behavior of osteosarcoma cells.Methods Recombinant plasmid with endostatin gene was amplified in vitro.The osteosarcoma cell line UMR116 cells were transfected by liposomes and were screened by using Zoecin.The proliferation capability of UMR116 cells was evaluated by MTT method;the secretion of endostatin was measured by ELISA,and the capability of invasion and mobilization of UMR116 cells were observed through Transwell loculus.Results The proliferation capability of transfected UMR116 cell was similar with those untransfected cells.Transfected cells could secrete endostatin with concentration of 350 ng/ml in the culture media.The capability of invasion and mobilization of UMR116 cells didn't change after transfection.Conclusion Osteosarcoma cells can be transfected with endostatin gene in vitro,without affecting the proliferation,mobilization,and invasion capability of cells.
