摘要
为研究丁型肝炎病人肝组织Fas/FasL和HDAg表达及意义,采用免疫组化单、双标记技术,检测了48例丁型肝炎病人肝组织Fas、FasL和HDAg表达,且以54例乙型肝炎作对照。结果发现,Fas以肝细胞浆表达为主,HDAg以肝细胞核和胞浆表达为主,二抗原表达及分布呈一致性。FasL以浸润淋巴细胞浆或肝细胞核表达为主,与HDAg表达及分布不全一致,3成分在各型肝炎的表达强度有显著性差别意义。提示:Fas/FasL和HDAg均与肝组织炎症活动和病理损害相关,HDV感染可诱导肝细胞表达Fas,CTL通过Fas/FasL途径介导的肝细胞凋亡与HDV致病可能有关。
To study the role and expression of Fas antigen(Fas Ag)、 Fas ligand (FasL) and hepatitis D virus antigen( HDAg) in liver tissue of the patient with hepatitis D. Forty eight liver biopsy samples from the patients with hepatitis D were studied using immunohistochemistry to determine Fas、FasL、HDAg.Fas/HDAg、 FasL/HDAg、Fas/FasL were double stained. The results showed that FasAg was mainly expressed in the cytoplasm of hepatocytes, and HDAg in the nucleus of hepatocytes, FasL in the cytoplasm of infilirating lyinphocytes or in the nucleus of hepatocytes. These positive cells were distributed among infiltrating lymophocytes at the periportal region especially at the advanceing edges of areas of “piecemeal necrosis”. The distribution and expression of Fas and HDAg was rather closed in hepatitis D. The degree of the expression of Fas、FasL and HDAg in hepatocytes had distinct difference in different hapatitis types( P <0.01).These suggest that the distribution and the expression of Fas、FasL and HDAg are significantly correlated with the activity of imflammation and the severity of the liver damage, and HDV infection may induce the expression of Fas in the hepatocytes, and apoptosis caused by cytotoxic T cells( CTL) through Fas FasL interaction may play an important role in the hepatocytes injury by HDV infection.
