协同指数法筛选银翘天甘组方的最佳剂量配比 被引量:14

摘要 为了研究银翘天甘组方中金银花、连翘、天花粉、甘草4味中药的最佳剂量配比,利用SI测定方法设计试验,微管-平板法测定各试验药物的MIC,计算SI值,依据呈现协同作用的SI值获得组方剂量配比,以浸膏率与MIC为验证依据,确定组方的最佳剂量配比。结果表明,四味中药配伍体外抗菌活性呈现协同作用的SI值在0.625~0.750之间,银翘天甘对大肠埃希菌有协同作用的剂量配比为2∶3∶9∶4,对猪链球菌2型有协同作用的剂量配比为1∶6∶72∶2;验证试验结果表明,前者浸膏率显著高于后者(P〈0.05),对大肠埃希菌的体外抗菌活性极显著高于后者(P〈0.01),MIC为20μg/mL,对猪链球菌2型的抗菌活性也优于后者,MIC为30μg/mL。由此得出银翘天甘组方中金银花-连翘-天花粉-甘草体外抗菌的最佳剂量配比为2∶3∶9∶4。 In order to study on the optimal dose ratio of Flos loniceraer,Forsythia fruit,Radix trichosanthis and Glycyrrhizae radixt in Yinqiaotiangan compound,the test was designed by the assay method of the synergic index(SI).The minimal inhibition concentration(MIC) of every drug was determined by the microbule-plate methods.SI was calculated by the formula.The dose matching of four Chinese herbs would come from SI of presenting synergism.Results showed that the SI of presenting synergism was 0.625-0.750.The results of MIC showed that the matching of Flos loniceraer,Forsythia fruit,Radix trichosanthis and Glycyrrhizae radixt was 2∶3∶9∶4 which presented the better antibacterial effect on E.coli in vitro.The results of MIC showed that the matching of them was 1∶6∶72∶2 which presented good antibacterial effect on Streptococcus suis Ⅱ in vitro.The results of verification test showed that extracted rate of the former was significantly higher than that of the latter(P0.05).The antibacterial effect of the former was significantly higher than that of the latter(P0.01) on E.coli and antibacterial effect of the former was better than that of the latter on Streptococcus suis Ⅱ in vitro.MIC of the former was 20 μg/mL to E.coli and 30 μg/mL to Streptococcus suis Ⅱ.So the optimal matching of Flos loniceraer,Forsythia fruit,Radix trichosanthis and Glycyrrhizae radixt in Yinqiaotiangan compound was 2∶3∶9∶4.
