大剂量卡铂对豚鼠前庭及耳蜗功能的影响 被引量:1

EFFECT ON COCHLEAR AND VESTIBULAR FUNCTIONS WITH CARBOPLATIN IN HIGH DOSES IN GUINEA PIGS

摘要 近年来,一些学者报道注射卡铂能够选择性地损伤灰鼠的内毛细胞,而外毛细胞则基本不受影响,并认为注射卡铂的灰鼠是选择性内毛细胞损伤的良好模型,而在豚鼠所做的有限的研究却众说不一。豚鼠是国内常用的耳科实验动物,为了明确卡铂对鼠豚是否存在耳毒性,是否能够利用注射卡铂的豚鼠进行与选择性内毛细胞损伤有关的动物实验研究。本研究采用大剂量(相当于国外灰鼠注射剂量的2倍)卡铂静脉注射观察豚鼠给药前后眼震电图(ENG)、听觉脑干电反应(ABR)、耳声发射(OAE)的变化。实验发现给药前后各项指标均无明显改变,与给药前相比,差异无显著性意义。表明卡铂不影响豚鼠前庭及耳蜗功能,对于卡铂的耳毒性,豚鼠与灰鼠之间可能确实存在种系差异。 In recent studies many scholars demonstrated that intravenous injection of carboplatin could induce extensive inner cell(IHC) lesions and minimal outer hair cell(OHC)damage in the cochlear of chinchilla Chinchilla was viewed as a perfect animal model for the study of selective IHC loss In the limited studies using the guinea pig as a model there were quite inconsistent results The purpose of this study was to confirm the ototoxic effects of carboplatin in high doses on the guinea pig and the necessity of animal studies for selective IHC loss A series of electrical physiological results were recorded using auditory brain stem evoked responses(ABR), otoacoustic emissions(OAE)and electronystagmogrphy(ENG) We discovered no statistically significant changes between pretreatment and posttreatment The study showed carboplatin in this animal didn′t affect cochlear and vestibular functions Interspecies variations exist with carboplatin in different models
