摘要
目的 建立一种简便易行的细菌 D N A 指纹图谱分析的方法用于医院感染流行病学研究。方法 选用肠道菌广泛存在的基因间重复片段作为引物,比较不同的聚合酶链反应( P C R) 的扩增条件,对临床分离的大肠埃希菌的 P C R 扩增产物电泳图谱作了初步分析。结果 建立了重复片段引物 P C R 的扩增方法,对大肠埃希菌扩增出了较为丰富的区带图谱,其中1 例病人的两个分离株其 P C R 产物图谱变化与其抗生素耐药变化极为相关。结论 该方法简便易行,适合于医院感染流行病学研究。
Objective To establish a convenient PCR method for bacterial DNA fingerprints analysis in epidermiology research of nosocomial infection. Methods By applying the widespread enterobacter repetitive intergenic consensus(ERIC) as a primer, we amplified DNA products from clinical isolated E.coli strains by rep PCR and compared the electrophoresis profiles with the antibiotic susceptability spectrum.Results Rep PCR was established, for creating useful DNA fingerprints of E.coli for epidermiology research.Two strains from one patient showed high relationships between PCR profiles and antibiotic resistance spectrums.Conclusion The method is practical and economical for epidermiology research in nosocomial infection.
