摘要 目的筛选拮抗FK506所致肾毒性的药物。方法将卡托普利、内皮素受体拮抗剂及钙通道阻滞剂3种药物分别与FK506一起进行大鼠离体肾灌流研究,观察3种药物对FK506致肾灌流阻力升高及肾小球滤过率下降的影响。结果卡托普利对肾灌流阻力及肾小球滤过率的变化无影响;内皮素受体拮抗剂能纠正肾小球滤过率的下降,钙通道阻滞剂既可纠正肾灌流阻力的上升也能纠正肾小球滤过率的下降。结论钙通道阻滞剂对FK506所致急性肾毒性有良好拮抗的作用。 Objective We have found FK506 has some toxic effects on kidneys. The present study was designed to identify the drug(s) counteracting the nephrotoxic effects.Methods Isolated rat kidneys were perfused with FK506 solely as control and in combination with separate captopril (Cap), endothelin receptor antagonist (ETRA) and calcium channel blocker (CaB) in experimental groups.Results Cap had no effects on reversing the increase of perfusion resistance (PR) and the decrease of glomerular filtration rate (GFR) induced by FK506, ETRA could partly reverse the GFR, while CaB reversed both the increase of PR and the decrease of GFR.Conclusion CaB has substantial effects preventing the development of FK506-induced nephrotoxicity.
