摘要
目的:探讨 C E A重组痘苗病毒对 C E A 阳性肿瘤的预防作用。方法:构建 C E A 阳性小鼠 Hepa 肝癌细胞模型( Hepa C E A+ ) 。将实验鼠分为4 组,先分别皮下接种痘苗病毒( 野生型 W V V 或重组型 C E Ar V)3 次,再分别颈背部皮下注射 C E A 阳性或 C E A 阴性同源鼠 Hepa 肝癌细胞。观察动物反应并在接种癌细胞后每周测量一次肿瘤大小。结果:3 个对照组各鼠颈背部皮下均可见有瘤块形成,并且以相近速率快速增长,3 组之间无统计学上的显著差异,而接种 C E Ar V/ Hepa C E A+ 组各鼠在观察期限内均无肿瘤形成。整个实验过程中实验鼠未表现有毒副反应。结论: C E Ar V 通过诱导机体的免疫系统产生 C E A 特异性的细胞和体液免疫应答反应,能有效预防小鼠移植性 C E A 阳性肿瘤的形成。
Objective:To study the preventive effect of CEA recombinant vaccina virus (CEA rV)on CEA positive tumor.Methods:The CEA positive cell model of liver cancer were constructed.Forty mice, which were divided into four groups,were vaccinated by subcutaneous injection with wild type vaccinia virus (WVV) or CEA rV for three times respectively at 14 day intervals and followed by subcutaneous injection of CEA positive or CEA negative liver tumor cells.The animal responses were observed and the subcutaneous tumors were measured weekly after tumor cells injected.Results:Subcutaneous tumors grew in all mice in three control groups,and grew rapidly at approximately the same rate (P >0.05),in contrast,in 6 weeks period, no tumor grew in the group Ⅳmice which CEA rV and CEA positive tumor cells were injected.During the whole testing period,no toxic or side effect was observed in all animals.Conclusions:Through inducing the body's immune response, CEA rV could effectively prevent the formation of murine CEA positive tumor transplanted.
