摘要
以优良玉米自交系昌7-2、齐319、郑58为试验材料,针对培养基类型、温度处理、激素配比等因素,优化玉米成熟胚再生体系。结果表明:玉米种子置于4℃预处理36 h,再置于34℃水浴中预处理36 h后愈伤诱导率有一定的提高。最适愈伤诱导培养基为NB+4 mg/L2,4-D+10 mg/LAgNO3,继代培养基为NB+2 mg/L 2,4-D+0.2mg/L 6-BA+10 mg/L AgNO3,分化培养基为NB+0.5 mg/L 6-BA。
The maize inbred lines Chang7-2, Qi319 and Zheng58 were used to establish the regeneration system and optimize the influence factors of mature embryos including the formula of medium and the temperature treatment. The results showed that the pretreatment on seed by 4℃for 36 h and 34℃for 36 h was beneficial to increase the rate of callus induction. The suitable induction medium was NB + 4 mg/L 2,4-D, the subculture medium was NB + 2 mg/L 2,4-D + 0.2 mg/L 6-BA + 10 mg/L AgNO3, and differentiation medium was NB + 0.5 mg/L 6-BA.
