摘要
从小麦粉中提取纯化了一种神经酰胺类物质,并对其结构进行了分析。实验以小麦粉为材料,首先用索氏提取法以95%乙醇为溶剂进行粗提,粗提物经石油醚萃取、旋转蒸发仪浓缩后,过三次硅胶柱层析提取,再经二次重结晶纯化,得到白色无定型粉末状神经酰胺类物质。使用高效液相色谱/蒸发光散射检测器(HPLC/ELSD)对其进行纯度检测,用红外光谱(IR)和质谱(MS)对其结构进行鉴定。实验结果分析表明该物质为分子量397.3,分子式C24H47NO3的神经酰胺类化合物。
A species of ceramide homolog from wheat powder was extracted and purified and its structure analysis was carried out. Wheat powder was treated with 95% of ethanol by means of sorbitic extraction at first. Then the crude product was extracted from petroleum ether,concentrated by rotatory evaporator,extracted three times by silica gel column chromatography,recrystallized twice,respectively,and white undefined ceramide powder was obtained. By means of HPLC/ELSD,the purity of the product was detected. Its structure was characterized via IR and MS spectroscopy. Experimental results showed that the molecular weight of the ceramide was 397.3,molecular formula was C24H47NO3.
