摘要
基于紫外分光光度法测定北五味子藤茎总木脂素含量,通过计算6种大孔树脂的吸附率和解析率,筛选出分离纯化总木脂素的最佳树脂为AB-8大孔树脂。研究各工艺因素对吸附和解析效果的影响,得到分离纯化总木脂素的最佳工艺为:总木脂素浓度为1.511 mg/mL时,最大上样量约为6倍柱床体积,洗脱流速1 mL/min,蒸馏水洗脱体积为8倍柱床体积,洗脱剂95%乙醇用量为5倍柱床体积。在此工艺条件下的洗脱率达到78.89%。
According to the UV spectrophotometry method for measuring the total lignans content from caulis of Schisandra chinensis,the author screens out the best resin for obtaining pure lignans by calculating the adsorption and desorption rate of six types of macro porous resins.With the evaluation of the dosage of loading buffer,velocity of adsorption,the volume of distilled water and alcohol in the eluent,the writer then comes out with the best purification process for obtaining total lignans content from AB-8 macro porous resins.As the experiment reveals,in order to obtain lignin with the density of 1.511 mg/mL,the maximum loading amount is approximately 6 times to the column volume as well as distilled water and 95% alcohol takes 8 and 5 the column volume,respectively.Also,the flow rate of elution is measured as 1 mL/min.In conclusion,with this advanced purification process,the elution rate can reach as high as 75.53%,providing us with a better way to purify lignans content from caulis of Schisandra chinensis and will be make good use in the industrial world in the future.
