摘要
目的:研究宫颈癌淋巴结转移的差异表达基因,探究宫颈癌淋巴结转移的分子机理,寻找可能用于早期诊断及临床治疗的分子靶标,探讨表达谱新篇在宫颈癌诊断中的应用价值;方法:冷冻微切割取3例病人癌旁上皮组织、癌组织、及转移淋巴结癌组织,提取总RNA,反转录,采用罗氏NimbleGen基因表达谱芯片筛查差异表达基因,利用GenCLip软件进行文献挖掘研究这些差异表达基因的功能关系。结果:癌组织和癌旁上皮组织相比有1444条基因上调;2108条基因表达下调;迁移淋巴结中癌组织和癌旁上皮组织相比有5313条基因上调;5169条基因表达下调;并对差异表达基因进行功能分析。结论:表达谱芯片可用来快速高通量筛查差异表达基因。
Objective:To study the differentially expressed gene in cervical cancer with lymph node metastasis,explore the molecular mechanism and the molecular pathogenesis,and to investigate potential gene markers in the diagnosis and treatment of the disease.Methods:Paracancerous epithelium tissue,tumor tissue and lymph node metastasis were gained from three patients.Total mRNA was extracted and synthesized by reverse transcription.The differentially expressed genes were screened by gene expression profile chip(NimbleGen),and the function was explored by GenCLip.Results:A total of 1 444 differentially expressed genes were up-regulated in tumor tissue as comparing with paracancerous epithelium,and 2 108 were down-regulated.While there were 5 169 up-regulated genes and 5 313 down-regulated genes in lymph node metastasis as comparing with paracancerous epithelium.Then functional analysis was also carried out.Conclusions:Expression gene profile chip is a powerful strategy to identify novel cancer associated genes,and it is also useful in rapidly exploring the roles of the novel genes.
