血管钠肽、C型钠尿肽和心房钠尿肽舒血管作用的对比 被引量:11

摘要 本实验采用离体血管灌流方法,观察和比较血管钠肽(VNP)、C型钠尿肽(CNP)和心房钠尿肽(ANP)对大鼠肺动脉、腹主动脉和腹腔静脉的舒张作用。结果表明,VNP、CNP和ANP对离体大鼠的保留内皮与去内皮的肺动脉、腹主动脉和腹腔静脉均有浓度依赖性舒张作用。对内皮完整的肺动脉、腹主动脉和腹腔静脉的最大舒张反应(Rmax),VNP分别为(76±17)%,(51±14)%和(62±14)%;CNP分别为(31±8)%,(22±7)%和(41±8)%;ANP分别为(38±10)%,(41±10)%和(11±4)%。内皮完整的肺动脉、腹主动脉和腹腔静脉最大舒张的半数有效浓度(EC50),VNP分别为16±11nmol/L,35±18nmol/L和12±8nmol/L;CNP分别为148±112nmol/L,299±84nmol/L和14±12nmol/L,;ANP分别为66±47nmol/L,16±15nmol/L和909±445nmol/L。VNP的舒张作用显著强于CNP和ANP,有显著差异(P<0.05~0.01)。但在保留内皮组与去内皮组间无显著差异(P>0.05)。松弛肺动脉的作用依次为VNP>ANP≥CNP;松弛腹主动脉的作用依次为VNP>ANP>CNP;松弛腹腔静脉的作用依次为VNP>CNP>ANP。以上结果提示,VNP的舒血管作用显著强于CNP和ANP,它是一个新合成的、强效的、非内皮依赖性的血管松弛多肽,较CNP和ANP更具有潜? The vasorelaxing effects of vasonatrin peptide (VNP), C-type natriaretic peptide(CNP) and atrial natriuretic peptide (ANP) on isolated rat pulmonary artery, abdominalaorta and celiac vein were measured by in vitro perfusion. The results showed that VNP,CNP and ANP caused concentraion-dependent relaxaion in isolated rat pulmonary artery,abdominal aorta and celiac vein with endothelium or without endothelium. The maximalresponses (Rmax) of VNP were (76±17)%,(51±14)% and (62±14)% inpulmonary artery , abdondnal aorta and celiac vein with endothelium respectively, whereasthose of CNP were (31±8)%, (22±7)% and (41±8)%, and ANP (38±10)%,(41±10)% and (11±4) %. The median effective concentration (EC50) of VNP were16±11, 35±18 and 12±8 umol/L in pulmonary artery, abdominal aorta and celiacvein with endoithelium respectively, while those of CNP were 148±112, 299±84 and 14±12 mnol/L, and ANP 66±47, 16±15 and 909±445 nmol/L. VNP were moreeffective than CNP and ANP, and the differences were statistically significant (P<0.05~0. 01). The potency of these peptides for relaxing the blood vessels can be summarizedas: VNP>ANP≥CNP for pulmonary nrtery; VNP>ANP>CNP for abdominal aorta;VNP>CNP>ANP for celiac vein. There was no significant difference between vesselswith intact endothelium and those denuded of endothelium (P>0. 05).
