摘要
目的研究顺铂腹腔热化疗对大鼠卵巢癌血清CA125表达水平的影响,观察它对卵巢癌的疗效,探讨其治疗卵巢癌的机理,从而为临床应用腹腔热化疗治疗卵巢癌提供实验依据。方法将体外培养的近交系Fischer344大鼠低分化上皮性卵巢癌细胞株NuTu-19,接种于雌性近交系Fischer344大鼠腹腔内,建立卵巢癌腹腔转移瘤模型;将模型动物随机分组,进行三个疗程的腹腔热化疗后处死大鼠。利用电化学发光免疫分析法(ECLIA)比较不同治疗组间血清CA125水平变化,观察不同治疗方法对卵巢癌的疗效。结果 1)所有实验动物均表现出不同程度的血性腹水及腹腔转移瘤微小结节病灶,经过3次IPHC治疗后,对照组大鼠体重和腹围仍呈增长趋势,各治疗组有不同程度下降,但各治疗组之间体重及腹围的变化并无显著差异(P>0.05);2)ECLIA对血清CA125测定结果发现:应用顺铂治疗的大鼠血清CA125表达水平较不加药组显著降低(P<0.05);不同加热方式对血清CA125表达水平的影响具有差异(P<0.05),一次性加热组和持续微波加热组均较不加热组表达降低,持续微波加热组血清CA125表达水平显著降低(P<0.05),本实验并未发现药物和加热方式对血清CA125表达水平的影响具有交互作用(P>0.05)。结论 1)腹腔热疗、化疗均能降低Fischer344大鼠低分化上皮性卵巢癌血清CA125表达水平;2)持续微波腹腔热化疗能较其他方法显著降低血清CA125表达水平,是一种理想的具有综合治疗效果的组配。
Objective To study the effects of intraperitoneal hyperthermic chemotherapy on cell proliferation of rat ovarian carcinoma.At the same time,we should observe the curative effect,and approach its mechanism in order to provide the experimental basis in clinical application of ovarian carcinoma.Methods NuTu-19,a cell line derived from a poorly differentiated adnocarcinoma of Fischer 344 rat,was cultured in vitro.To establish the animal models of peritoneal carcinomatosis of inbred line rat Fischer 344.To execute the rats after 3 IPHC.Electrochemiluminescence immunization and innunohistochemistry were respectively to detect the level of CA125 in blood serum.Results(1)All the rats were displayed with inequality hemorrhagic ascites and metastatic tumor focus of infection in abdominal cavity.After 3 IPHC,the weight and abdomen circumference in the control group were still increasing,while the therapy groups presented inequality decrease.But their variation in all therapy groups had no statistical significance(P〈0.05).(2)The results of ECLIA were as follows.The groups of using cisplatin had a lower level of CA125 in blood serum(P〈0.05);the different heating models presented significant difference(P〈0.05),and the groups of persistent heating by microwave was the lowest(P〈0.05).These two factors did not display interaction in this experiment(P〈0.05).Conclusions(1)Both the intraperitoneal thermotherapy and the intraperitoneal chemotherapy can reduce the level of CA125 in blood serum of the poorly differentiated adenocarcinoma of Fischer 344 rat.(2)The IPHC with persistent heating by microwave can make decrease of the level of CA125 further in blood serum,which is the optimum combination in this experiment.
