Encapsulation Efficiency and Drug Loading Determination of β-elemene in β-elemene-Polybutylcyanoacrylate Nanoparticles by RP-HPLC

摘要 目的:建立β-榄香烯聚氰基丙烯酸正丁酯纳米粒(β-ELE-PBCA-NP)包封率和载药量的测定方法。方法:采用低温超速离心法,RPHPLC法测定包封率和载药量,色谱柱为Diamonsil TMC18(150mm×4.6mm,5μm),流动相为乙腈-水(90:10),检波长为210nm。结果:β-榄香烯在(22.4~179.2)μg·ml^(-1)(r=0.9996)的范围内呈现良好线性关系,平均加样回收率为96.9%,RSD为1.7%,测得的平均包封率为90.17%,平均载药量为7.07%。结论:本方法简便、准确,可用于β-ELE-PBCA-NP包封率和载药量的测定。 Objective: To develop a RP-HPLC method for the encapsulation efficiency(EE) and drug loading(DL) determination in β-elemene-Polybutylcyanoacrylate nanoparticles(β-ELE-PBCA-NP).Methods:By Ultracentrifugation at low temperature,determinate encapsulation efficiency and drug loading of β-elemene-Polybutylcyanoaerylate nanoparticles by RP-HPLC.The analysis was performed on Diamonsil TMC18 (150mm×4.6mm,5μm),The mobile phase was aeetonitrile-water (90:10),The wavelength of the detector was 210nm. Results:β-elemene presented a good linear correlation in range of (22.4~179.2)μg·ml-1 (r=0.9996). The average recovery was 96.9%(RSD=1.7%).The average EE of β-ELE-PBCA-NP is 90.17%,and the average DL is 7.07%. Conclusion:The method is simple and accurate,and suitable for the EE and DL determination of β-ELE-PBCA-NP.
