摘要
根据高致病性猪繁殖与呼吸综合征病毒(highly pathogenic porcine reproductive and respiratory syndrome virus,HP-PRRSV)的Nsp2基因部分缺失的特征,设计合成1对特异性引物,对长春某猪场4份疑似HP-PRRSV感染病死猪的淋巴结、脾脏、肺脏等病料进行RT-PCR检测,结果均扩增出226 bp的特异片段,与预想结果一致。取病死猪淋巴结、肺脏、脾脏等组织,用10%中性福尔马林固定,石蜡包埋,HE染色,进行病理组织学分析,结果显示,淋巴结和脾脏中淋巴细胞严重坏死,肺脏呈弥漫性间质性肺炎病变,肝脏、肾脏、脑等器官实质细胞均出现不同程度的损伤。
According to the highly pathogenic porcine reproductive and respiratory syndrome virus(HP-PRRSV) Nsp2 gene partially missing features,designed and synthesized a pair of specificity primers,4 dead pigs' lymph nodes,spleens,lungs that suspected HP-PRRSV infection were detected by RT-PCR,226 bp specific segments were amplificated and agreed with the desired results.The lungs,spleens,lymph nodes were taken and fixed in 10% formalin,the paraffin section were made to observe the change of this abnormal tissue,the results showed that the lymphocytes in lymph nodes and spleen were serious necrosis,lung was diffuse interstitial pneumonia,the substantial cells were damaged of different degree in liver,kidney and brain.
