摘要
目的:探讨不同孵育浓度和不同光照剂量密度喜泊芬介导的光动力治疗(photodynamic therapy,PDT)对人食管癌细胞Eca-109体外效应的影响。方法:以不同浓度喜泊芬孵育食管癌Eca-109细胞,并分别在不同光照剂量密度(0、12、24、30J/cm2)下行PDT,24h后通过四甲基偶氮唑盐(MTT)法检测细胞生存率。结果:不同孵育浓度喜泊芬在4种不同光照剂量密度下食管癌Eca-109细胞生存率间均有显著差异(P<0.01)。在同一光照剂量密度下,不同喜泊芬浓度的食管癌Eca-109细胞的生存率有显著差异(P<0.01),而同一喜泊芬孵育浓度下,不同光照剂量密度的食管癌Eca-109细胞的生存率有显著差异(P<0.01)。结论:喜泊芬的不同孵育浓度和不同光照剂量密度对人食管癌Eca-109细胞体外PDT效应有显著影响。
Objective:To investigate the influence of HiPorfin mediated photodynamic therapy(PDT) effect with different incubation concentration and under different laser doses in human esophageal cancer cell line Eca-109 in vitro.Methods:Human Eca-109 cells were incubated in vitro with HiPorfin in different incubating concentration,and followed by irradiation in different laser doses(0,12,24,30 J·cm-2,respectively).Cell survival rate of each concentration was measured by 3-2,5-diphenyltertrazolium bromide(MTT) assay 24 hours after PDT.Results:The cell survival rate between different HiPorfin incubating concentration and different laser doses were all significantly different(P0.01).While under the same laser doses,the cell survival rate was also significantly different with different incubating concentration(P0.01).And while under the same concentration,the cell survival rate was also significantly different with different laser doses(P0.01).Conclutions:The incubating concentration and laser doses of HiPorfin significantly influence photodynamic therapy effect in human esophageal cancer cell line in vitro.
