摘要
以白粉桃茎段为材料,通过根癌农杆菌介导用反义多聚半乳糖醛酸酶(PG)基因对桃进行了遗传转化.将预培养3d的受体材料经根癌农杆菌菌液(D600=0.4)浸染10min后共培养60h,转移至分化培养基MS+1.0mg·L-16-BA+0.5mg·L-1NAA+50mg·L-1卡那霉素+250mg·L-1头孢氨苄青霉素上诱导产生不定芽,将抗卡那霉素的不定芽先后在含有50mg·L-1卡那霉素的生长、增殖和生根培养基上继续选择,获得了完整植株,经PCR检测初步证明反义PG基因已导入桃中.
Using the peach stem as material,transgenic peach was obtained transformed with antisense PG gene by Agrobacterium tumefaciens.The receptor materials were pre-cultured for 3 d,infected 10 minutes in A.tumefaciens (the D600=0.4 ),and co-cultured for 60 h,then transferred to differentiation medium MS +1.0 mg·L-16-BA +0.5 mg·L-1NAA +50 mg·L-1 km +250 mg·L-1 cef to induce shoots.The kanamvcin-resistant shoots were further selected on the media containing 50 mg·L-1 km,the media where the shoots grew,multiplicated and rooted,the whole plants were obtained.The PCR test preliminaly showed that the antisense PG gene has been introduced into the plants.
