人参皂苷Rb1对吗啡慢性作用SK-N-SH细胞增殖及[Ca^(2+)]_i的影响 被引量:2

摘要 目的:探讨人参皂苷Rb1对吗啡慢性作用SK-N-SH细胞增殖及[Ca2+]i的影响。方法:采用四甲基偶氮唑盐比色法(MTT)研究人参皂苷Rb1对吗啡慢性作用SK-N-SH细胞增殖的影响,应用荧光分光光度法观察人参皂苷Rb1对吗啡慢性作用SK-N-SH细胞内[Ca2+]i的影响。结果:与对照组相比,吗啡慢性作用48h可显著抑制SK-N-SH细胞的增殖,细胞内游离钙离子浓度显著增高(P<0.01);单独加入16μmol.L-1Rb1可显著促进SK-N-SH细胞的增殖(P<0.05);人参皂苷Rb1(16μmol.L-1,32μmol.L-1)对细胞进行预处理能缓解吗啡对细胞的抑制作用(P<0.05)。单独加入16μmol.L-1Rb1可使SK-N-SH细胞内[Ca2+]i显著降低(P<0.01);慢性吗啡作用SK-N-SH细胞48h,能使细胞内[Ca2+]i明显升高(P<0.01);慢性吗啡作用SK-N-SH后,加入(16μmol.L-1,32μmol.L-1)人参皂苷Rb1能有效地抑制吗啡引起的细胞内[Ca2+]i升高(P<0.01)。结论:人参皂苷Rb1可显著缓解吗啡对SK-N-SH细胞的抑制作用和吗啡引起的[Ca2+]i升高。 Objective:To explore the effects of ginsenoside-Rb1 on cell multiplication and [Ca2+]i in the SK-N-SH treated with chronic morphine.Methods:MTT was used to research the effects of ginsenoside-Rg1 on cell multiplication in the SK-N-SH cells treated with chronic morphine.Fluorospectrophotometry was used to observe the effects of ginsenoside-Rb1 on [Ca2+]i in the SK-N-SH cells treated with chronic morphine.Results:Morphine significantly inhibited cell multiplication and resulted in calcium overload in the SK-N-SH cells treated with chronic morphine for 48 h(P0.01).Adding 16 μmol·L-1 ginsenoside-Rb1 could enhance cell multiplication(P0.05).However ginsenoside-Rb1(16 μmol·L-1,32 μmol·L-1)could relieve the depressant effect of morphine on cell multiplication(P0.05).Adding 16 μmol·L-1 ginsenoside-Rb1 made [Ca2+]i significantly lower in SK-N-SH cells(P0.01).Cell [Ca2+]i significantly increased(P0.01)when stimulated by morphine.Ginsenoside-Rb1(16 μmol·L-1,32 μmol·L-1)could significantly inhibit morphine induced calcium overload(P0.01).Conclusion:Ginsenoside-Rb1 can significantly relieve the depressant effect of morphine on cell multiplication and elevate the morphine induced [Ca2+]i in the SK-N-SH cells.
