摘要
目的:建立液相色谱-串联质谱法测定血浆中曲马多、杜冷丁和美沙酮的含量。方法:血浆中的杜冷丁、曲马多和美沙酮用乙醚提取,采用Zorbax SB-C18(2.1 mm×50 mm,3.5μm)柱子,柱温35℃,流速:0.40 ml/min,流动相为乙腈-0.1%甲酸。ESI(电喷雾离子源),正离子检测,选择离子监测(SIM)方式进行定量分析。结果:杜冷丁、曲马多和美沙酮的浓度在4 ng/ml~800 ng/ml范围内线性良好(r?0.997),平均萃取回收率在76%以上,日内、日间精密度RSD?13%。结论:该方法的样品预处理快速简便,检测专一灵敏,可满足曲马多,杜冷丁,美沙酮临床研究药物浓度测定的要求。
Objective:To establish a LC-MS method for determination of meperidine,tramadol and methadone in human plasma.Methods:Meperidine,tramadol and methadone were extracted from plasma samples with ethylether and separation was carried out on Zorbax SB-C18 column(2.1 mm×50 mm,3.5 μm) at 35℃,acetonitrile-0.1% formic acid was used as mobile phase with the flow rate of 0.40 ml/min.Electrospray ionization ion(ESI) source was applied and operated in positive ion mode,and selective ion monitoring(SIM) mode was used to quantify meperidine,tramadol and methadone.Results:The calibration curve in the concentration range of 4 ng/ml~800 ng/ml showed a good linear(r≧0.997) for meperidine,tramadol and methadone.The average extraction recoveries were better than 76%.Intra-day RSD and inter-day RSD were less than 13%.Conclusion:This method is specific,sensitive and accurate,and it can be used to detect meperidine,tramadol and methadone in forensic and clinical analytic toxicology.
