高效液相电化学联用测定尿甲氧基肾上腺素和甲氧基去甲肾上腺素 被引量:3

Determination of metanephrine and normetanephrine in urine by HPLC-ECD

摘要 目的:建立了高效液相电化学(HPLC-ECD)联用测定尿甲氧基肾上腺素(M)和甲氧基去甲肾上腺素(NM)的方法。方法:利用固相萃取柱提取尿甲氧基肾上腺素(M)和甲氧基去甲肾上腺素(NM),并用高效液相电化学(HPLC-ECD)联用测定其含量。结果:尿甲氧基肾上腺素(M)和甲氧基去甲肾上腺素(NM)的回收率分别为90%~102%和89%~94%,色谱分离效果好,批内(M)相对标准偏差为5.3%,(NM)为3.2%。(M)的批间相对标准偏差为10.3%,(NM)的批间相对标准偏差为7.8%。正常人与嗜铬细胞瘤患者的含量差别明显(P<0.01)。结论:该方法操作简单,结果可靠,可用于对嗜铬细胞的临床诊断。 Objective:To establish a method for determination of metanephrine and normetanephrine in urine by HPLC-ECD.Methods: Metanephrine and normetanephrine were extracted from urine by solid phase extraction column and detected by HPLC-ECD.Results: The recoveries were in the range of 96%~102% for metanephrine(M) and 89%~94%for normetanephrine(NM)respectively.The separation effect of chromatographic was good.The relative standard deviation within batch and between batch for(M) was 5.3% and 10.3% and for(NM)was 3.2% and 7.8% repectively.Normal person and pheochromocytoma patient′s with significantly different levels of clinical significance(P0.01).Conclusion: This method is simple and result is reliable,can be used for clinical diagnosis of pheochromocytoma.
