摘要
以艳丽花楸的侧芽和种子萌发苗为外植体,MS为基本培养基,添加不同配比的植物生长调节剂,进行离体培养。结果表明:较之种子萌发苗,当年侧芽外植体更适合作为诱导材料。在芽的诱导及增殖阶段,当激素水平为6-BA0.5 mg.L-1+NAA0.10 mg.L-1时,增殖效果较好,分化率可达90%,单芽平均分化芽数大于5个。在生根诱导阶段,采用质量浓度为2.8 mg.L-1的IBA较为合适,生根率可达到95%。
Lateral buds and seedlings of Sorbus decora were cultured for callus induction on MS medium supplemented with different hormone combinations.The results showed that the 1-yr-old lateral bud explants was a more suitable material than seedlings.The optimal hormone concentration for the proliferation and propagation was 6-BA0.5 mg·L-1+NAA0.10 mg·L-1,and the differentiation rate reached to 90%,with more than 5 buds differentiated from single bud on average.The optimal rooting medium was 2.8 mg·L-1of IBA,and the rooting rate reached to 95%.
