高效液相色谱法测定湿疹乳膏中氯霉素和醋酸曲安奈德含量

Determination of Chloramphenicol and Triamcinolone Acetonide Acetate in Shizhen Cream by HPLC

目的建立测定湿疹乳膏中氯霉素和醋酸曲安奈德含量的高效液相色谱法。方法色谱柱为Inertsil ODS-SP柱(150mm×4.6mm,5μm),以甲醇-水-乙醚(62:38:2)为流动相,流速1.0mL/min,检测波长为320nm和240nm。结果醋酸曲安奈德质量浓度在10.2～34.0μg/mL范围内与峰面积线性关系良好,r=0.9999(n=6),平均回收率为99.12%,RSD为1.39%(n=6);氯霉素质量浓度在99.3～496.5μg/mL范围内与峰面积线性关系良好,r=0.9999(n=6),平均回收率为97.03%,RSD为1.75%(n=6)。结论所用方法定量准确、操作简便,可作为湿疹乳膏的质量控制方法。

Objective To establish an HPLC method for the determination of chloramphenicol and triamcinolone acetonide acetate in Shizhen Cream.Methods The Inertsil ODS-SP column was used with methanol-water-ether（62：38：2） as the mobile phase.The flow rate was 1.0 mL /min and the detecting wavelengths were 320 nm and 240 nm.Results The linear range was 10.2-34.0 μg /mL（r = 0.999 99,n = 6） and 99.3-496.5 μg /mL（r = 0.999 9,n = 6） for triamcinolone acetonide acetate and chloramphenicol respectively.The recovery rate and RSD were 99.12%,1.39%（n = 6） and 97.03%,1.75%（n = 6） respectively.Conclusion The method is accurate in quantitation,and easy and simple to be operated,which can be used as the quality control method of Shizhen Cream.
