摘要
目的优选岗梅根中总皂苷的最佳提取工艺条件。方法采用正交设计L9(34)方法,以齐墩果酸为对照品,通过比色法测定总皂苷的含量,考察乙醇用量、乙醇体积分数和提取时间等因素对提取率的影响。结果水浴提取法的最佳工艺为16倍量40%的乙醇提取1 h,在此条件下,总皂苷的质量分数达到1.355%;超声提取法的最佳工艺为16倍量40%的乙醇提取40 min,在此条件下,总皂苷的质量分数达到1.706%。超声法优于水浴法。结论优选的最佳提取工艺条件,为开发岗梅根的有效部位提供一个科学、合理的理论依据。
Objective To optimize the extraction condition for total saponins from Roughhaired Holly Root.Methods The orthogonal test L9(34) was adopted with oleanolic acid as control and the content of total saponins detected by colorimetric method as index for investigating the effects of ethanol amount,ethanol concentration and extraction time on the extraction rate.Results The optimal technology of water bath extraction was extracting with 16 times amount of 40% ethanol for 1 h.Under this condition,the mass fraction of total saponins reached to 1.355%.The optimal technology of ultrasonic wave extraction was extracting with 16 times amount of 40% ethanol for 40 min.Under this condition,the mass fraction of total saponins reached to 1.706%.The ultrasonic extraction method was superior to the water-bath extrac-tion.Conclusion Optimizing the extraction technology may provide the scientific and reasonable basis for developing the active parts from Roughhaired Holly Root.
