摘要
目的探讨赖氨酰氧化酶(LOX)在不同侵袭、转移潜能的乳腺癌细胞株的表达及LOX与MMP-2、MMP-9的关系。方法以RT-PCR检测LOX mRNA在乳腺癌细胞株MDA-MB-231、MCF-7的表达水平。40只裸鼠随机分为2组,每组裸鼠的乳腺脂肪垫内分别接种上述不同的细胞,建立人乳腺癌原位移植瘤模型。6周后测定肿瘤体积,SP免疫组织化学法检测移植瘤中LOX、MMP-2、MMP-9的表达情况,并对数据进行统计学处理。结果高侵袭、转移潜能的乳腺癌细胞株MDA-MB-231中LOXmRNA表达量显著高于低侵袭、转移潜能的乳腺癌细胞株MCF-7(P<0.01);裸鼠接种癌细胞后第6天均有肿瘤形成,至6周时MDA-MB-231组肿瘤体积为(758.84±241.08)mm3,MCF-7组为(210.36±101.06)mm3,差异有统计学意义(P<0.01)。LOX与MMP-2、MMP-9的表达呈正相关(P<0.01)。结论乳腺癌细胞高侵袭、转移的潜能可能与LOX、MMP-2、MMP-9的高表达及其相互之间协同作用相关。
Objective To explore the expression of lysyl oxidase(LOX)in breast cancer and its relation to MMP-2 and MMP-9.Methods The expression level of LOX mRNA was detected by RT-PCR in breast cancer cell lines MDA-MB-231 and MCF-7 firstly.BALB/c nude mice were randomly divided into two groups,20 mice in each group.MDA-MB-231 and MCF-7 cells were implanted into the mammary fat pad of mice in each group respectively.Tumor volume was estimated six weeks after cell inoculation.Immunohistochemical method S-P was used to detect the expressions of LOX,MMP-2 and MMP-9 in the transplanted tumors,and the data were analyzed statistically.Results The highly invasive/metastatic breast cancer cell line MDA-MB-231 had much higher expression level of LOX mRNA than that of MCF-7(P0.01).Tumor occurred six days after cell inoculation.At the 6th week after inoculation,the tumor average size in group MDA-MB-231 and group MCF-7 were(758.84±241.08)mm3 and(210.36±101.06)mm3 respectively,and the difference between the two groups was statistically significant(P0.01).LOX was positively correlated with MMP-2 and MMP-9(P0.01).Conclusions The highly invasive/metastatic potential of breast cancer may be related to the high expression levels of LOX,MMP-2 and MMP-9,and to their synergistic effect.
