NQO1蛋白在家族聚集性肝癌中的表达及临床意义

Expression and clinical significance of protein NQO1 in hepatocellular carcinoma-clustered families

目的研究NQO1蛋白在家族聚集性肝癌中的表达以及探讨其与临床病理关系。方法运用免疫组化SP法检测41例非家族聚集性肝癌组织(非家族癌组)及其癌旁组织、34例家族聚集性肝癌组织(家族癌组)及其癌旁组织、32例良性病肝组织NQO1蛋白表达的情况。并分析NQO1蛋白表达与肝癌临床病理因素间的关系。结果 NQO1蛋白在肝癌组织以及正常肝组织中均有表达,家族癌组以及非家族癌组中肝癌组织NQO1蛋白表达均低于正常组(P<0.05)。家族癌组该蛋白表达比非家族癌组更低(P<0.05)。家族癌组中肝癌组织NQO1蛋白表达明显低于癌旁组织(P<0.05)。非家族癌组中肝癌组织NQO1蛋白表达也明显低于癌旁组织(P<0.05)。家族癌组、非家族癌组癌旁组织的NQO1蛋白表达均低于正常肝组织(P<0.05)。家族癌组比非家族癌组中癌旁组织的NQO1蛋白表达更低(P<0.05)。家族聚集性肝癌中NQO1蛋白表达与年龄、性别、HBsAg和病理分级无关,而与AFP定量、肿瘤大小以及家族中患癌个体数有关。结论 NQO1蛋白表达的差异可能在家族聚集性肝癌的发生或发展中起一定作用,这对家族聚集性肝癌的预防、早期诊断和早期治疗有极其重要的临床价值。

Objective To study the expression of protein quinone oxidoreductase 1（NQO1）in hepatocellular carcinoma（HCC）-clustered family,and to investigate its correlation with clinicopathological parameters.Methods By immunohistochemistry S-P method,the expression levels of protein NQO1 were examined in HCC tissues and the adjacent liver tissues of 41 and 34 HCC-patients that were from the families without clustered-HCC（non-family cancer group）and the families with clustered-HCC（family cancer group）respectively.32 cases of normal liver tissues were used as control.The relationship between expression of NQO1 and clinicopathological parameters of HCC was analyzed.Results The expression of NQO1 existed in both HCC tissues and in normal liver tissues,but the expression level in HCC tissues was lower than that in normal liver tissues（P0.05）.Family cancer group had lower NQO1 expressions in HCC tissues and HCC-adjacent liver tissues than those of non-family cancer group（P0.05）.Both non-family cancer group and family cancer group had lower expression of NQO1 in HCC-adjacent liver tissues than that in normal liver tissues（P0.05）.The expression of NQO1 in familial cancer group was irrelevant with age,sex,HBsAg and HCC histopathology grade,while was correlated with AFP lever,tumor size and number of HCC patient in family history.Conclusions NQO1 may play an important role in development of family clustered-HCC.The expression of protein NQO1 has extremely important clinical value in prevention,early diagnosis and treatment of family clustered-HCC.
