摘要
目的:探讨刺蒺藜血清对黑素瘤细胞增殖及黑素合成的影响。方法:体外培养小鼠B16黑素瘤细胞,制备大鼠含药血清,分别采用四甲基偶氮唑蓝比色法(MTT法)和NaOH裂解法检测细胞增殖及黑素含量。结果:刺蒺藜对黑素细胞增殖及黑素含量具有低剂量(10g)抑制、高剂量(30g)促进作用(P<0.01),中剂量(20g)对细胞增殖及黑素含量的影响倾向于促进作用,但与空白对照组比较无统计学意义。结论:刺蒺藜对黑素细胞增殖及黑素含量的影响与剂量有关,具有低浓度抑制,高浓度激活的双向调节作用。
Objective To observe the effects of serum containing Tribulus terrestris on proliferation and melanogenesis in B16 melanoma cells.Methods B16 melanoma cells was cultured in vitro.Serum containing Tribulus terrestris was prepared.MTT and NaOH method were used to detect proliferation and melanogenesis respectively.Results The serum containing Tribulus terrestris increased the amount of melanin at higher concentrations(30g) but acted to the reverse effect at lower concentrations(10g)(P0.01).Compared with blank control group,The amount of melanin at middle concentrations(20g) had no significant difference(P0.05).Conclusions the effects of serum containing Tribulus terrestris on proliferation and melanogenesis in B16 melanoma cells correlated with the concentration of Tribulus terrestris.It presented a two-way regulation on proliferation and melanogenesis in cultured B16 melanoma cells.
