坐骨神经横断后远期体外培养的施万细胞形态及其分泌BDNF和NT3功能的变化

Long-term changes of morphology of in vitro cultured Schwann cells in distal stump of sciatic nerve following denervation and their ability of secreting BDNF and NT-3

摘要 目的观察坐骨神经横断后体外培养的远端施万细胞(SCs)形态及其分泌脑源性神经营养因子(BDNF)和神经营养因子3(NT3)功能的变化。方法依据坐骨神经横断时间,16只SD大鼠分为横断后1个月组(A1组,3只)、2个月组(A2组,5只)和3个月组(A3组,6只);C组(2只)为正常对照组。取各组横断坐骨神经远端进行SCs培养。免疫荧光染色观察细胞形态;ELISA检测SCs分泌BDNF、NT3水平。结果坐骨神经横断后SCs形态均发生改变,以A1组最为明显。与C组比较,A1组SCs分泌的BDNF增多(P<0.05);A2、A3组BDNF和NT3均较A1、C组明显减少(P<0.05);但A2组与A3组间各指标无统计学差异(P>0.05)。结论坐骨神经横断损伤1个月后,其远端SCs形态发生改变;2个月后BDNF和NT3分泌量下降。 Objective To investigate the long-term changes of morphology of in vitro cultured Schwann cells(SCs) from transected distal stump of sciatic nerve and their ability of secreting brain-derived neurotrophic factor(BDNF) and neurotrophin 3(NT-3).Methods The SCs were obtained from the distal stumps of sciatic nerves following transected for different periods of time and cultured.Sixteen SD rats were divided into groups of A1 (denervated for 1 month, 3 rats), A2 (denervated for 2 months,5 rats), A3 (denervated for 3 months,6 rats) and rats in group C (2 rats) were sham operated as the controls.The morphology of SCs was observed by immunofluorescence staining and the levels of BDNF and NT3 secreted by SCs were assessed by ELISA.Results Morphological changes of SCs were observed,especially in group A1.Compared with group C,BDNF secreted by SCs in group A1 was increased (P0.05).The levels of BDNF and NT3 were significantly lower in groups of A2 and A3 than those in groups of A1 and C(P0.05).No significant differences in BDNF and NT3 were seen between group A2 and group A3(P0.05).Conclusion There are changes in morphology of SCs from the distal stumps of transected sciatic nerves.BDNF and NT3 secreted by SCs are decreased in 2 to 3 months following transection.
