熊果酸对胃癌细胞中cofilin-1蛋白表达的影响 被引量:1

摘要 目的研究熊果酸作用胃癌细胞BGC-823后丝切蛋白cofilin-1表达的变化。方法熊果酸作用于细胞BGC-823,双向电泳结合质谱检测胞质内cofilin-1的表达,免疫印迹检测cofilin-1蛋白和细胞色素C的表达。结果胞质内cofilin-1蛋白表达降低(P<0.01),而线粒体表达却明显升高(P<0.01),同时细胞色素C在胞质中的表达量也明显下调(P<0.01)。结论熊果酸作用BGC-823,可能使cofilin-1蛋白从胞质转位至线粒体内,促使细胞色素C释放,激活caspase级联反应,诱导细胞凋亡。 Objective To investigate expression changes of cofilin-1 in cell BGC-823 treated with ursolic acid. Methods BGC-823 cell was treated with ursohe acid, expression changes of eofilin-1 was detected by 2-D-electrophoresis and mass spectrum, protein expression of cofilin-1 and cytochrome C was detected by Western blot. Results Lower expression of cofilin-1 in eytoplasm(P 〈0. 01 ) , but obvious raise in mitochondria ( P 〈 0. 01 ), and down- regulation of cytochrome C in eytoplasm(P 〈 0. 01 ) were found. Conclusion BGC-823 treated with ursolic acid may transfer cofilin-1 transposition from cytoplasm to mitochondria, which promoted eytochrome C released from mitochondria, activates caspase cascade, and induces cell apoptosis.
