载万古霉素硅溶胶缓释系统的体外抑菌研究 被引量:3

摘要 目的制备载万古霉素硅溶胶(vancomycin-sol,VCM-SOL)涂层缓释系统,探讨其体外抑菌性能。方法采用正硅酸乙酯酸化水解法及溶胶-凝胶法制备载万古霉素(VCM)浓度为20 mg/ml的VCM-SOL涂层钛钢片作为实验组,普通钛钢片作为对照组。利用红外光谱分析系统(FT-IR)来分析实验组的表面基团;选用金黄色葡萄球菌(staphylococcus aureus,S.A)作为实验菌,通过细菌计数进行洗脱液抑菌实验,扫描电镜检测钛钢片表面细菌黏附实验,对比观察实验组和对照组的体外抗菌效果。结果硅溶胶对万古霉素有良好的吸附作用,实验组的洗脱液相对于对照组明显抑制了金黄色葡萄球菌的生长;实验组表面黏附的细菌较为分散,且大部分金黄色葡萄球菌被分解,而对照组表面细菌结构正常,细菌集结成团,易形成生物膜。结论万古霉素以硅溶胶为载体携载到钛合金表面涂层中后,具有良好的体外抗菌活性。 Objective To prepare vancomycin-sol coating and investigate its bacteriostasis characteristics in vitro.Methods VCM20-SOL coating which contains 20 mg/ml vancomycin was prepared by acid-catalyzed tetraethylorthosilane with sol as carrier.Then a titanium alloy sheet was coated with the prepared VCM20-SOL coating with sol-gel technique,and the blank titanium alloy sheet served as the control group.Fourier transform infrared spectrum system(FT-IR) was used to analyze the surface groups of the VCM20-SOL coated titanium alloy sheet,Staphylococcus aureus(S.aureus) was employed as test bacterium.The antibacterial effect of the eluent from the experimental sheet and the control group was observed.Bacterial adhesion of the experiment and control sheets were evaluated.Results The silica sol had good properties in adsorption of vancomycin in vitro.The eluent of the experimental sheet obviously inhibited the growth of S.aureus when compared with that of control group.The bacteria adhered to the surface of experimental sheet was in dispersion,and the most of S.aureus were decomposed.However,the structure of the bacteria on the control group surface was normal,and they adhered to agglomerate,and were easy to form biofilm.Conclusion Our prepared vancomycin-sol coating has good antibacterial activity in vitro.
