胃泌素和5-羟色胺在海洛因戒断、复吸大鼠胃窦的表达 被引量:1

Expressions of gastrin and 5-HT in gastric antrum of rats going through heroin withdrawal then relapse

摘要 目的探讨海洛因戒断、复吸对大鼠胃窦内胃泌素(gastrin,Gas)、5-羟色胺(5-hydroxy tryptamine,5-HT)阳性细胞的影响。方法成年雄性SD大鼠35只,按配对原则随机分为正常对照组(n=5)、生理盐水对照组(n=15)和实验组(n=15),实验组大鼠又分为海洛因戒断组、美沙酮脱毒治疗组和海洛因复吸组,每组5只。采用免疫组织化学SABC法及图像分析法,观察Gas、5-HT在大鼠胃窦中表达的变化。结果①在海洛因戒断、复吸期间,Gas、5-HT在胃窦表达增强,与正常对照组及生理盐水对照组比较,戒断及复吸组大鼠胃窦内的Gas、5-HT阳性细胞免疫染色加深,平均灰度值均低于正常及盐水对照组,且阳性细胞计数增多,差异有统计学意义(P<0.05)。②脱毒治疗组与正常及生理盐水对照组比较,大鼠胃窦内的Gas、5-HT阳性细胞的免疫染色强度无明显变化,平均灰度值及细胞计数差异无统计学意义(P>0.05)。结论在海洛因戒断、复吸期间,胃窦通过分泌Gas、5-HT增多的方式,参与了机体对抗海洛因的损伤;美沙酮脱毒治疗后胃窦分泌Gas、5-HT有所减少,可能与胃窦功能逐步趋于正常有关。 Objective To explore the expressions of gastrin(Gas) and 5-hydroxy tryptamine(5-HT) in the gastric antrum of rats who go through heroin withdrawal and then relapse.Methods A total of 35 adult male SD rats were randomly divided into 3 groups,normal control group(n=5),saline control group(n=15) and experiment group(n=15).Experiment group was subdivided into heroin withdrawal group(n=5),methadone detoxication group(n=5) and heroin relapse group(n=5).All animals from experimental group were given injections of 3 mg/kg heroin at the first day,twice per day,then with an increase of 3 mg/kg every day for another 8 d and then followed a boost injection of 0.8 mg naloxone to establish heroin withdrawal model.Then the left 10 rats of the experimental group received a continued decrease injection of methadone for 6 d(5,4,3,2,1 and 0.5 mg/kg) to establish methadone detoxication rats.The left 5 rats were injected with heroin again from 9 to 3 mg/kg for 7 d with the dose decreased every day by 1 mg/kg.All rats from experimental group were sacrificed after the establishment of corresponding models.Immunohistochemical SABC and image analysis were performed to detect the expressions of Gas and 5-HT.Results The expressions of Gas and 5-HT were increased during heroin withdrawal and relapse in rat antrum.Compared with normal control and saline control,heroin withdrawal and relapse group had more intense immunohistochemical staining,lower mean grey scale and more Gas and 5-HT positive cells(P0.05).There was no significant difference in the mean grey scale and positive cells counts of Gas and 5-HT(P0.05) in methadone detoxication group when compared with normal and saline control.Conclusion Gas and 5-HT show a trend of increased expression in rat antrum during withdrawal,and withdrawal and relapse,suggesting that they participate in the anti-heroin process.After methadone detoxication,the expressions of Gas and 5-HT were close to normal levels,which indicating the function of antrum turns to normal.
