摘要
本试验筛选出诱导川贝母鳞茎的最佳外植体。选取生长不同时期的川贝母植株鳞茎外植体分别接入由不同激素浓度配比的培养基中,在不同培养条件下获得川贝母鳞茎快速增殖的最佳培养条件,并检测出再生鳞茎的总生物碱含量。结果表明:开花期鳞茎为鳞茎增殖的最佳外植体,其最佳培养基配方为MS+6-BA 2.0 mg.L-1+NAA 0.2 mg.L-1,川贝母出苗期、开花期和果实期植株鳞茎外植体再生鳞茎的总生物碱含量分别为0.180%,0.190%,0.182%,均高于野生川贝母鳞茎的总生物碱含量0.109%。
In this trial,the best explant for the induction of Fritillariae cirrhosae bulb was selected.Different growth periods of Fritillariae cirrhosae bulb explants were planted into culture media with different hormone concentration combination,respectively.The best culture conditions for rapid proliferation of Fritillariae cirrhosae bulb were got and the total alkaloid content in the regenerated bulb was detected.The result showed that flowering stage bulbs was the best explants for the proliferation of bulb.The optimum culture medium was MS +6-BA 2.0 mg·L-1 + NAA 0.2 mg·L-1.The total alkaloid content in the seedling stage,flowering stage and fruit period of regenerated bulb from Fritillariae cirrhosae bulb explants were 0.180 %,0.190 %,0.182 %,respectively,higher than that of the wild Fritillariae cirrhosae bulb which was 0.109 %.
