摘要
研究了不同浓度TDZ对两个非洲紫罗兰品种Elizabeth和Nilson Blue叶片外植体愈伤组织形成、类型以及体细胞胚形成与发育的影响。结果表明,Elizabeth与Nilson Blue在培养反应、再生潜力及形态发生途径上均存在显著差异。TDZ提高叶片愈伤率,促进愈伤组织生长,适宜浓度为0.5~1.0 mg.L-1;TDZ与NAA组合能高效诱导Elizabeth胚性愈伤组织形成和体细胞胚发生。扫描电镜观察表明:随TDZ浓度增加,体细胞胚发育加快,同步化程度高。Elizabeth叶片外植体体细胞胚诱导最适培养基为MS+0.2 mg.L-1NAA+1.0 mg.L-1TDZ。
Effect of TDZ on formation and types of calli,induction and development of somatic embryos on leaf explants of 2 African violets varieties,Elizabeth and Nilson Blue were studied.The result showed that there were significant differences on response of culture regeneration potential and approach of morphogenesis between Elizabeth and Nilson Blue.TDZ increased the rate of callus formation and promoted the growth of calli with a suitable range of 0.5-1.0 mg·L-1.By combining with NAA,TDZ induced embryogenic callus formation and somatic embryogenesis actively.Scanning electron microscopy(SEM) observation indicated that TDZ prompted the development of somatic embryos with increasing of TDZ concentration,thus improved synchronization level of somatic embryos.MS + 0.2 mg·L-1 NAA + 1.0 mg·L-1 TDZ was the best somatic embryo induction medium for leaf explants of Elizabeth.
