摘要
本研究以中国美利奴羊(新疆军垦型)为实验材料,采用PCR-RFLP技术和测序分析KAP1.3基因遗传多态性。结果表明:KAP1.3基因经限制性内切酶HapⅡ酶切,得到3种基因型,分别为AA型、BB型和AB型,其中AA型频率为0.7550,BB型频率为0.0741,AB型频率为0.1709,等位基因A频率为0.8405,等位基因B频率为0.1596;KAP1.3基因经限制性内切酶AdeI酶切得到CC型频率为0.6586,DD型频率为0.1248,CD型频率为0.2166,等位基因C频率为0.7668,等位基因D频率为0.2331。证明KAP1.3基因多态性丰富,能够用于优质细毛羊毛性状功能基因的研究。
PCR-RFLP and sequencing analysis were used to identify polymorphisms of KAP1.3 gene in Chinese Merino(XinJiang JunKen Type).The results showed KAP1.3 gene had three genotypes with HapⅡ digestion,AA,BB and AB individual.AA genotype frequency was 0.7550,BB genotype frequency was 0.0741,AB genotype frequency was 0.1709.A allele frequency was 0.8405,B allele frequency was 0.1596.KAP1.3 gene had three genotypes with AdeI digestion.CC genotype frequency was 0.6586,DD genotype frequency was 0.1248,CD genotype frequency was 0.2166,C allele frequency was 0.7668,D allele frequency was 0.2331.It was showed there were abundant genetic polymorphism of KAP1.3 gene.KAP1.3 gene could be used to study functional gene of wool trait in high quality merino sheep.
