摘要
首次开展附子种胚培养技术体系研究。以吸水膨胀的种胚为外植体,筛选获得最佳诱导愈伤组织培养基MS+2,4-D 1mg/L,愈伤组织分化出苗培养基MS+6-BA4 mg/L+NAA0.05 mg/L,增殖培养基MS+6-BA4 mg/L+NAA0.3 mg/L,增殖倍数可达20倍,生根培养基1/2MS+IBA1 mg/L+KT1 mg/L,移栽基质以蛭石∶珍珠岩∶泥炭=1∶1∶1为最佳,成活率可达80%。
To study the embryo culture method of Aconitum camichaeli Debx.for the first time.Swollen embryo was taken as explants.The best callus inducing medium was screened out as MS+2,4-D 1 mg/L.The best seedling differentiation medium was screened out as MS+6-BA 4 mg/L+NAA 0.05 mg/L.The best proliferation medium was screened out as MS+6-BA 4 mg/L+NAA 0.3 mg/L and the proliferation multiples reached 20.The best rooting medium was screened out as 1/2MS + IBA 1 mg/L + KT 1 mg/L.The best transplanting substrate was screened out as the mixture of vermiculite,perlite and turves with equivalent volume.Survival rate of transplanting culture seedling reached 80 %.
