全缘冬青及其变型变种亲缘关系RAPD和AFLP分析

摘要 采用RAPD和AFLP技术对8个全缘冬青及其变型变种基因组进行扩增,以研究其物种间亲缘关系及遗传多样性。结果表明:在RAPD分析中,20条引物共扩增出187条多态性条带,多态率为60.75%。在AFLP分析中,10对选择性引物组合共扩增出350条谱带,其中268条具有多态性,占76.57%。综合RAPD和AFLP技术的聚类结果,"小叶枸骨叶"全缘冬青和"小叶密节"全缘冬青亲缘关系较近,全缘冬青和全缘冬青"卵叶密节"亲缘关系最近。这为冬青属植物的杂交育种与种质创新提供理论依据。 RAPD and AFLP techniques were used to amplify the genomic DNA of 8 Ilex integra Thunb. species and its forma and varieties to study the genetic relationship and species genetic diversity. The results showed that 187 polymorphic bands with the polymorphism rate at 60.75% were obtained using 20 primers by RAPD analysis. Rich polymorphic bands were amplified both from 10 selective AFLP primer pairs in AFLP analysis and 350 bands were obtained,of which 268(76.57%) were polymorphic. Integrating the cluster results of RAPD and AFLP techniques,it was found that 'Xiaoyegouguye' Ilex integra and 'xiaoyemijie' Ilex integra had the nearest relationship,Ilex integra and 'Luanyemijie' Ilex integra had a near relationship. This results provide theoretical basis for cross breeding and germplasm enhancement of Ilex L. tree species.
