摘要
目的:探讨不同治疗时间法舒地尔(fasudil)对心力衰竭大鼠心肌细胞内钙离子浓度的影响。方法:将升主动脉缩窄术后心力衰竭Wistar雌性大鼠随机分为4组,每组10只,两组为心力衰竭组,给予0.9%NaCl溶液,0.1ml,腹腔注射,每日2次;另两组给予fasudil5mg/kg,腹腔注射,每日2次(n=10);分别治疗2周或6周。同时制备两组机械性套环模拟组模型作为对照组(n=10),不予处置。观察每组大鼠各项指标变化。结果:心力衰竭鼠心肌细胞钙离子浓度([Ca2+]i)显著增高,fasudil组随干预时间增加改善心力衰竭症状更明显,但心肌细胞内[Ca2+]i无明显改变。结论:法舒地尔能随干预时间的延长而有效的缓解心力衰竭症状,但不降低细胞内[Ca2+]i,可能为一种新的、有效的治疗心力衰竭的血管扩张剂。
Objective:To investigate impact of calcium concentration in myocardial cells of the rat with heart failure in treatment with fasudil in different treatment time.Methods:Female Wistar rats after ascending aorta coarctation operation were divided into four groups,10 every group,two groups of heart failure were treated with the physiological saline,0.1 ml,abdominal injection,2 times per day;two groups were treates with fasudil 5 mg/kg,abdominal injection,2 times per day(n=10);treatment of two weeks or six weeks.At the same time produced a set of machinery two analog groups model as a control group(n=10) and did not be disposed.The indexes of all groups were observed.Results:Calcium concentration in myocardial cells of the rat with heart failure([Ca2+]i) had a significant increase,with the intervening period increasing,heart failure symptoms of fasudil group improved more obvious,but the heart muscle cells [Ca2+]i had no significant change.Conclusion:With the intervening period increasing,the symptoms of heart failure is effectively alleviate,but cells [Ca2+]i is not lower,it could be a new,effective treatment of heart failure in the expansion of blood vessels.
