摘要
为了准确测定铜绿假单胞菌发酵液中鼠李糖脂的量,研究了蒽酮-硫酸法测定发酵液中鼠李糖脂的反应条件,并对测定结果的稳定性、精密度、回收率等进行了考察.结果表明:(1)蒽酮硫酸法能够测定发酵液中鼠李糖脂的量.空白对照采用未接种的发酵液而不是蒸馏水,消除了其他成分的干扰.(2)该方法稳定性RSD=0.79%,重复性RSD=1.9%,回收率=99.98%,精密度RSD=0.1%,方法学考察合格.(3)按照该方法测得发酵液中鼠李糖脂的量为1 057.75 mg/L.采用蒽酮硫酸法测定发酵液中鼠李糖脂,方法可靠可行.
In order to determine the amount of rhamnolipid in the zymotic fluid of Pseudomonas aeruginosa accurately,the reaction conditions of rhamnolipid in the zymotic fluid were studied by the method of anthrone-sulfuric acid,and the stability,precision and recovery rates of the results were also investigated.Results:(1)The amount of rhamnolipid in the zymotic fluid could be measured by the method of anthraquinone-sulfuric acid.Unvaccinated zymotic fluid instead of distilled water was used in the blank control,which could eliminate the interference of other ingredients.(2)The stability RSD of the method was 0.79%,repeatability RSD 1.9%,recovery rate 99.98%,precision RSD = 0.1%.Therefore,the investigation of its methodology was up to standard.(3)According to that method,the amount of rhamnolipid measured in the zymotic fluid was 1 057.75 mg/L.Meantime,it was reliable and feasible to use the method of anthrone-sulfuric acid to determine the rhamnolipid in the zymotic fluid.
