摘要
目的:研究金属硫蛋白对活性氧诱导的血管平滑肌细胞增殖的影响。方法:用过氧化氢和甲萘醌分别作为内源性和外源性活性氧刺激平滑肌细胞增殖,应用细胞总蛋白定量,3HTdR 参入,细胞周期时相测定等方法测定细胞增殖。结果:过氧化氢(25 μmol·L- 1)和甲萘醌(0 .5 μmol·L- 1) 可诱导培养的大鼠主动脉平滑肌细胞增殖,使其蛋白总量增加,S期细胞比例增高,DNA合成率增加。金属硫蛋白可以抑制过氧化氢和甲萘醌诱导的血管平滑肌细胞增殖。结论:金属硫蛋白对预防由活性氧诱导的血管平滑肌细胞增殖所引起的疾病可能具有重要作用。
Objective: To investigate the effect of metallothionein on the proliferation of vascular smooth muscle cells induced by reactive oxygen species. Methods: Menadione and H 2O 2 were added to the medium of cultured rat aortic vascular smooth muscle cells as internal and extraneous reactive oxygen species respectively. Protein quantitative assay, 3H TdR incorporation and cell cycle phase analysis were used to detect proliferation of vascular smooth muscle cells. Results: The results showed that menadione(0.5 μmol·L -1 ) and H 2O 2 (25 μmol·L -1 ) enhanced the proliferation of vascular smooth muscle cells with protein concentration increased, and the ratio of S phase became higher and 3H TdR incorporation increased. Metallothionein could inhibit the effects of menadione and H 2O 2 on the proliferation of vascular smooth muscle cells. Conclusion: Metallothionein may have certain effect in preventing disease caused by reactive oxygen species induced vascular smooth muscle cells proliferation.
