摘要
采用活体内生长 SP2 / 0骨髓细胞为融合伙伴 ,用 SP2 / 0细胞与甲胎蛋白 (AFP)免疫的 BALA/ C小鼠脾细胞进行融合 ,建立了 2株稳定分泌抗 AFP单克隆抗体 (Mc AB)的小鼠杂交瘤株 (HMA2 15和HMA62 )。冻存两年后复苏 ,仍稳定地分泌特异抗体。培养上清的抗体效价达 10 -3 ,腹水效价达 10 -6 ,HMA2 15和 HMA64识别抗原表位不同 ,HMA2 15和 HMA64均属于小鼠 Ig G1亚类。结果表明两株细胞系能稳定地分泌抗人 AFP单抗 ,可为诊断和治疗的研究提供新的材料。
