摘要
目的研究乳腺癌组织中Raf激酶抑制蛋白(Raf kinase inhibitor protein,RKIP)、核转录因子-κBp65(nuclear factor-kappaB p65,NF-κBp65)蛋白的表达,探讨其表达与乳腺癌发生、发展的相关性。方法应用免疫组织化学染色方法(S-P法)检测54例乳腺癌组织、22例癌旁乳腺组织、14例乳腺增生症组织中RKIP、NF-κBp65蛋白的表达,并进行统计学分析。结果乳腺癌组织与癌旁乳腺组织、乳腺增生症组织中RKIP表达的阳性率为51.9%,81.8%,100%,乳腺癌组织与两者比较有显著性差异(P<0.05);NF-κBp65表达的阳性率分别为42.4%、13.6%、7.14%,与两者比较有显著性差异(P<0.05)。乳腺癌RKIP的表达与腋窝淋巴结转移相关(P<0.05),NF-kBp65的表达与乳腺癌的组织学分级、TNM分期显著相关(P<0.05)。RKIP与NF-κBp65在乳腺癌中表达负相关。结论 RKIP可能是NF-κBp65的一个重要调节因子,两者可能共同参与了乳腺癌的发生、发展过程。
Objective To investigate the expression and the interaction of Pokemon and NF-κBp65 in the course of carcinogenesis and metastasis of breast cancer.Methods The expression of RKIPand NF-κBp65 was examined in 54 cases of primary breast cancer,22 cases of adjacent noncancerous breast tissueand 14 cases of hyperpiasia of mammary glands tissues by immunohistochemical assay.Results The positive expression rate of RKIP was significantly lower in breast carcinoma tissues than that in adjacent noncancerous breast tissues or hyperpiasia of mammary glands tissues(P0.05).and the rates were 51.9%,81.8% and 100%,respectively.The positive expression rate of RKIP was significantly lower in breast carcinomas with axillary lymph node metastasis than that in breast carcinoma without axillary lymph node metastasis(P0.05).The positive expression rates of NF-κBp65 in breast carcinoma tissues,adjacent noncancerous breast tissues and hyperpiasia of mammary glands tissues were 42.4%,13.6%and 7.14%,respectively.Significant difference was observed between the positive expression rate of NF-κBp65 in breast carcinoma tissues and that in adjacent noncancerous breast tissues and in hyperpiasia of mammary glands tissues(P0.05).The expression of NF-κBp65 in breast carcinoma has a significant correlation with histological grading,TNM staging and axillary lymph node metastasis(P0.05).There was negative correlation between the expressions of RKIP and NF-κBp65 in breast carcinoma.Conclusion It is possible that RKIP locates the upstream of NF-κBp65 and plays an important role as a regulator of NF-κBp65.RKIP and NF-κBp65 may be implicated in the carcinogenesis of breast carcinoma together.
