摘要
目的建立反相高效液相色谱法(RP-HPLC)测定患者血浆中丙泊酚浓度。方法使用Phenomenex Gemini C18柱,(4.6 mm×250 mm,5μm),以甲醇-水(80∶20)为流动相,流速1.0 mL.min-1,柱温30℃,荧光激发波长(Ex)274 nm,发射波长(Em)312 nm。以甲醇沉淀法处理血浆,测定患者血浆中药物浓度。结果丙泊酚总浓度在0.108~8.64μg.mL-1与峰面积线性关系良好,低、中、高3个浓度方法回收率在95.5%~102.9%,日内RSD为2.5%~3.9%,日间RSD为3.9%~5.8%。结论本方法简便、快速,灵敏度高,适用于临床上丙泊酚血药浓度监测。
Objective To develop a reversed phase high-performance liquid chromatographic(RP-HPLC) method for the determination of propofol concentration in human plasma.Methods The determination was performed on Phenomenex Gemini C18(4.6 mm×250 mm,5 μm)column.The mobile phase consisted of methanol-water(80∶20) at 1.0 mL·min-1,the coloum temperature was 30℃,and the fluorescence wavelength Ex=274 nm,Em=312 nm.Methanol was used to precipitate blood samples to determine the drug concentration.Results The calibration curve of propofol concentration in human plasma was linear within 0.108-8.64 μg·mL-1.The method recoveries were 95.5%-102.9%,intra-day and inter-day RSDs were 2.5%-3.9% and 3.9%-5.8%,respectively.Conclusion The method is simple,rapid,sensitive,and suitable for clinical monitoring of propofol.
